An In Vitro Test for Drug Resistance in Haemonchus contortus

چکیده

A cambendazole-resistant (CR) strain of Haemonchus contortus, the large stomach nematode of sheep, had similar in vitro growth characteristics compared to the parent strain (BPL). When the in vitro cultivation system was modified to detect drug resistance: (1) exsheathed infective larvae of the BPL strain did not develop in a culture medium with a concentration of 2.5 /ug/rnl thiabendazole (TBZ), but the CR strain developed to the mid-4th larval stage in 6 days at the same concentration of TBZ; and, (2) larvae of both strains of//, contortus were killed prior to or in the 3rd molt in a culture medium with 2.5 /ug/ml levamisole.

Study of β-tubulin gene polymorphisms in Haemonchus contortus isolated from sheep populations in Khouzestan, southwestern Iran

In this report, β-tubulin gene polymorphism was investigated in Haemonchus contortus  populations isolated from sheep flocks in different regions of Khouzestan province, southwestern Iran. The samples were chosen on the basis of regional variation in benzimidazole treatment background of sheep flocks. Our objective was to study the relationship between treatment background andβ -tubulin gene po...

Immunodiagnosis of Haemonchus contortus infection in sheep by indirect enzyme linked immunosorbent assay (ELISA)

Indirect plate enzyme-linked immunosorbent assay was standardized and evaluated for its effectivenessin immunodiagnosis of haemonchosis in experimental and clinical cases in sheep by using somatic wholeadult antigen of H. contortus. Plate ELISA was standardized using 5 μg/well antigen concentration with1:100 and 1:1000 of sera and conjugate dilution. Indirect plate ELISA was able to demonstrate...

Haemonchus contortus Acetylcholine Receptors of the DEG-3 Subfamily and Their Role in Sensitivity to Monepantel

Gastro-intestinal nematodes in ruminants, especially Haemonchus contortus, are a global threat to sheep and cattle farming. The emergence of drug resistance, and even multi-drug resistance to the currently available classes of broad spectrum anthelmintics, further stresses the need for new drugs active against gastro-intestinal nematodes. A novel chemical class of synthetic anthelmintics, the A...

Comparison of constitutive and thiabendazole-induced expression of five cytochrome P450 genes in fourth-stage larvae of Haemonchus contortus isolates with different drug susceptibility identifies one gene with high constitutive expression in a multi-resistant isolate

Benzimidazoles (BZs) remain amongst the most widely used anthelmintic drug classes against gastro-intestinal nematode infections, although their efficacy is increasingly compromised by resistance. The primary underlying mechanisms for BZ resistance are single-nucleotide polymorphisms (SNPs) in the isotype 1 β-tubulin gene causing the substitutions F167Y, E198A or F200Y. However, resistance is b...
